Title:New Strategies for Post-Synthetic RNA Modifications

Abstract: RNA modifications play a crucial role in RNA biology and hold significant promise for therapeutic applications. However, key challenges remain in the field of RNA biomedical research, including RNA stabilization, targeted delivery, and site-selective modification of long RNA. Our lab is focused on developing innovative chemical strategies to address these hurdles. In this talk, I will present an "install-deliver-release" approach for RNA delivery, along with a catalytic method for site-selective RNA modification. Our work utilizes post-synthetic RNA acylation and disulfide exchange chemistry to enable efficient RNA conjugation and delivery. I will discuss thiol-mediated RNA delivery through a non-endocytic pathway, as well as GalNAc-mediated strategies for cell-selective RNA delivery. Finally, I will introduce a programmable and broadly applicable platform for site-selective RNA modification and highlight its potential applications.
